Isolation of Splenic B Cells: This section describes the negative-selection procedure used to isolate mouse splenic B cells and compares yields obtained from two laboratories.

Cell Characterization:  Specific subpopulation, developmental stage, and activation status of the isolated splenic B cells  characterized by multiparameter FACS analysis are shown.

Short-Term Cell Culture:  Conditions used to maintain short-term cultures of splenic B cells are described, including the selection of a suitable serum-free medium.

Functional Responses:  The functional integrity of our B cells is shown through measurements of ligand-induced changes in intracellular concentrations of Ca2+ and cyclic AMP and phosphorylation of specific sites within a panel of signaling proteins.
